Why a very small utility from TuneUp, is much speeder than defraggler?

The TuneUp program you mentioned looks fine if you want to pay for it, I guess.
CCleaner & a defragmentation program would much of the equivalent functions.
MyDefrag is a great program. I particularly use Defraggler when I want to target only those files which our fragmented.
They are complimentary programs but Defraggler focuses on informing you of the specific files that are fragmented.
Personally I would continue using MyDefrag as your primary defragmenter unless are looking for something else.

You should stick to one defrag program. Often the optimization routines are not exactly the same. Using a second program may therefore undo some of the optimizations that the first program did.

Defraggler is quick if you do a quick defrag, but the full defrag does both files & free space.
A lot of "fast" defraggers will perhaps just be doing a "quick" defrag instead of complete.
